چکیده

Purpose: To determine the level of proteolytic activity in lens epithelium samples from cataract surgery and to investigate a possible relation to the type of cataract. We wanted to estimate G-independent as well as Ca-dependent activity. Another purpose was to me.asure the activity of calpastatin, an endogenous inhibitor of calpain, which is a Ca-activated protease. Methods: Cataracts were typed and graded previous to surgery by slit lanm biomicroscouv. Caosulorhexis were immediatelv Dut in a hvootonic buffer containing i&h4 I?DTA and then frozen at-80%. After th'a~ing, the samples were incubated for one hour at room temperature, which lead to detachment of the epithelial cells from the capsule and rupture of the cell membranes, as confirmed by light microscopy. The solution of epithelial cell cytoplasma was then incubated with a synthetic fluorogenic substrate; N-succinyl-Leu-Tyr-7-amido4methylcoumain, in the absence and in the presence of calcium (5mM EDTA and 2mM Ca respectively). Caipastatin is a heat stable protein and its activity was measured as the inhibitory effect of the boiled sample solution on puritied rabbit lung calpain. Acavities were related to the level of LDH (lactat dehydrogenas) activity. i?esuuS: The means for the activities measured in the absence and presence of calcium appeared to be the same. However, there was .a big variation in the ratio of EDTA-/Ca-activity, although in most cases activity measured in the absence of Ca was slightly higher. Calpastatin activity was detectable in most samples. Conclusions: Net activity of C&dependent proteases seems to be inferior to total proteolytic activity. However, some samples do show a high Ca-dependent activity, but further analyses must be performed to be able to connect this to special diagnostic groups. Adhesion of lens capsule to intraocular lenses of polymethylmethacrylate, silicone, and acrylic foldable materials: an experimental study.

AIMS To investigate the adhesion characteristics of several intraocular lenses (IOLs) to the simulated and rabbit lens capsule. METHODS Adhesive force to bovine collagen sheets was measured in water with polymethylmethacrylate (PMMA), three piece silicone, and acrylic foldable IOLs. In rabbit eyes, phacoemulsification and IOL implantation were performed. Three weeks later, adhesion between th...

Refractive Change Caused Silicone Oil Adhesion to the Intraocular Lens Following Nd:YAG Posterior Capsulotomy

A 32-year-old female who had undergone the silicone oil removal procedure presented with visual disturbance in her left eye. Several months previous, she had cataract surgery in a private clinic, and a month later she had a Nd:YAG laser procedure for posterior capsulotomy. The slit-lamp examination revealed silicone oil droplets that had adhered to the intraocular lens where the posterior capsu...

Comparison of PMMA, foldable silicone and foldable acrylic hydrophobic intraocular lenses in combined phacoemulsification and trabeculectomy.

PURPOSE To compare the postoperative results of phacotrabeculectomy with implantation of PMMA, foldable silicone or foldable hydrofobic acrylic intraocular lens (IOL). SETTING Glaucoma unit, The Royal Liverpool University Hospital, Liverpool, United Kingdom. METHODS We studied a total of 124 eyes of three consecutive groups of patients with glaucoma and cataract that underwent phacotrabecul...

Lens epithelial cell regression on the posterior capsule with different intraocular lens materials.

BACKGROUND/AIMS Posterior capsular opacification (PCO) is caused by proliferation and migration of lens epithelial cells (LECs) across the posterior capsule and is the commonest cause of reduced vision after cataract surgery. The influence of intraocular lens (IOL) material on the process of LEC migration was studied. METHODS 90 eyes underwent standardised extracapsular surgery, with capsulor...

Silicone oil-intraocular lens interaction: which lens to use?

AIM To determine a suitable intraocular lens for implantation in patients at high risk of lens exposure to silicone oil in their lifetime. METHODS PMMA, AcrySof, AR40, AQUA-Sense, and Raysoft lenses were examined. Each lens was immersed for 5 minute intervals in balanced salt solution (BSS), in stained silicone oil, and again in BSS before being photographed in air and in BSS. Percentage sili...

Quantitative comparison of posterior capsule opacification after polymethylmethacrylate, silicone, and soft acrylic intraocular lens implantation.

OBJECTIVE To quantitatively compare the extent of posterior capsule opacification (PCO) after polymethylmethacrylate (PMMA), silicone, and soft acrylic intraocular lens implantation. PATIENTS AND METHODS A total of 240 eyes from 240 patients undergoing implant surgery were randomized into 3 groups based on the type of lens implanted: PMMA, silicone, and soft acrylic. The density value of PCO ...
